Heat shock

• Add plasmid to E coli and leave on ice for 10mins

• Place in a waterbath at 42^oC for 45s (shock)

• Add 0.5ml media and grow in incubator at 37^oC for 30-60min (1-2 reproductive cycles). Place plate in incubator at same time in order to dry: warm, dry plates increase the efficiency of transformation.

• Plate out cells and incubate overnight at 37^oC

For reasonably competent cells, use 1-2ul miniprepped plasmid to 50ul cells. If cells are less competent, more DNA may be required.
