Plasmid amplification

To make more copies of a plasmid, use a cloning strain of E. coli.

• Add a single colony of bacteria from a fresh plate (<1 week old) to 2-5ml of LB media (+ antibiotic) in a 50ml falcon tube

• Grow overnight in a shaker at 37^oC

• Pellet the cells by centrifuging at 3600rpm for 10mins. Discard the supernatent as biohazard waste.

The pellet can be frozen at -20^oC for future use. Alternatively, use immediately for miniprep DNA extraction using the kit from Qiagen. The Qiagen Spin Columns Handbook is uploaded to the DNA Biochemistry page.
