DNA

There are a selection of vectors available in the lab for subcloning including His and GST tags, and a number of different fusion proteins for solubility or expression screening (eg NusA, emGFP). The central stock is in the large -20 freezer, in the box marked Wallace Group plasmid stocks and is catalogued in a Google docs spreadsheet.

Information sheets on a large number of the plasmid sequences and cloning sites are uploaded below.

The plasmid stocks box is also the place to deposit a copy of any of your plasmids that you think someone else may be interested in. This is for labelled, sequenced plasmids which express protein only, not for a copy of everything that you've cloned in the hope it may work one day. Please update the Google docs spreadsheet when you deposit a plasmid.

The primers box is the place to deposit primers which might be useful to others in the group, or for long term storage. Please update the Google docs spreadsheet when you deposit a primer.

To determine the concentration of DNA

Double stranded DNA:

concentration in ng/ul = A₂₆₀ x dilution factor x 50

Single stranded DNA:

concentration in ng/ul = A₂₆₀ x dilution factor x 33
